Propagation of orchids using protocorm-like bodies (PLBs): A review

Abstract
Protocorm-like bodies (PLBs) represent a critical advancement in orchid
micropropagation, enabling mass production of genetically uniform plants for
commercial floriculture and conservation. This review synthesizes current
knowledge on PLB-based propagation systems across diverse orchid genera,
examining explant sources, culture conditions, hormonal requirements, and
emerging technologies. PLBs can be induced from various explant types including
shoot tips, leaves, protocorms, and roots. Cytokinins, particularly thidiazuron
(TDZ) and 6-benzyladenine (BA), are essential for PLB induction, while auxins
regulate subsequent development. Advanced systems including thin cell layer
culture, temporary immersion bioreactors, and optimized light regimes have
significantly enhanced propagation efficiency, with some protocols achieving
over 20,000 plantlets annually from minimal starting material.
